Title: Correlation of ET-1、VEGF-D with lymphangiogenesis and prognosis of laryngeal carcinoma
Abstract: OBJECTIVE To investigate the relationship between the expression of ET-1 and VEGF-D and lymphangiogenesis,lymph node metastasis, prognosis in patients with laryngeal cancer.METHODS The expression of ET-1 and VEGF-D in 45 specimens of the laryngeal cancer and 20 specimens of polyps of vocal cord tissues was observed by immunohistochemistry and real-time fluorescent quantitative PCR.LVD was evaluated by 5'-Nucleotides staining.MVD was evaluated by immunohistochemistry with the specific antibody CD34.The correlation analysis was made between ET-1,VEGF-D and clinical parameters.RESULTS The expression of ET-1,VEGF-D were higher in tumor tissue than in the polyp of vocal cord tissue.The expression of ET-1 was significantly correlated with peritumoral and intratumoral LVD and MVD,TNM stage,lymphatic metastasis and lymphovascular invasion.ET-1 was positively correlated with VEGF-D.And the expression of ET-1~+NEGF-D~+had inverse correlation with survival rate. CONCLUSION ET-1 overexpression could promote the lymphangiogenesis and lymph node metastasis of laryngeal carcinoma through upregulation of VEGF-D.Combined detection of the expression of both ET-1 and VEGF-D may be helpful in judging prognosis of laryngeal cancer.
