Title: Expression of COX-2 and MMP-2 in human pancreatic cancer and clinical significance
Abstract: Objectives:To investigate the relationship between the expression of cyclooxygenase-2 (COX-2) and matrix metallo- proteinase-2(MMP-2) and to define their association with clinicopathological characteristics of human pancreatic cancer. Methods: The expression of COX-2 and MMP-2 were detected by immunohisto- chemistry (two-pace Elivision method) in 33 cases of human pan- creatic ductal adenocarcinoma and 8 normal pancreas tissues. To analyze the relation between the expression of COX-2 and MMP-2 and the clinicopathological characteristics. Results: The high COX-2 expression ratio was 69.7%(23/33) in human pancreatic ductal adenocarcinoma. The high MMP-2 expression ratio was 63.6%(21/33). Both COX-2 and MMP-2 show negative in normal pancreas. High COX-2 and MMP-2 expression correlated lymph nodal metastasis , and COX-2 expression correlated with local tumor invasion (P= 0.020)but MMP-2 expression was not significant with local tumor invasion(P=0.163) .Both COX-2 and MMP-2 were not sig- nificant with age, gender or grade of differentiation (P0.05). A positive correlation was found between the expressions of COX-2 and MMP-2 (χ2=11.81,P0.005,r=0.598).Conclusions: Positive expressions of MMP-2 and COX-2 were increased in pancreatic can- cer, COX-2、MMP-2 expression correlated with lymph nodal metastasis ,and COX-2 expression correlated with local tumor invasion. COX-2 and MMP-2 may play a positive role to each other in the mechanism of lymph nodal metastasis in human pancreatic cancer.
