Title: Expressions and correlation analysis of CDK8 and MMP-7 in colorectal cancer tissues
Abstract: Objective To detect the expressions of cyclin-dependent proteinkinase 8(CDK8) and matrix metalloproteinases 7(MMP-7) in colorectal cancer tissues, and investigate their correlation and relationship with clinicopathologic features of colorectal cancer patients. Methods Immunohistochemical(IHC) method and RT-PCR were used to detect CDK8 and MMP-7 protein and mRNA expressions in tumor tissues,tissues adjacent to cancer from colorectal cancer patients and normal tissues..The correlation between CDK8 and MMP-7 expressions and relationship of CDK8 and MMP-7 expressions with clinicopathologic characteristics were analysized. Results Both protein and mRNA expressions of CDK8 and MMP-7 in tumor tissues were significantly higher than those in tissues adjacent to cancer and normal tissues(P 0.05)..Both protein and mRNA expressions of CDK8 were positively correlated with MM P-7expressions respectively(P 0.05). CDK8 Expression was significantly correlated with tumor infiltrating degree, Duke's stages, lymph node and distant metastasis(P0.05), but not with age, sex, tumor location and differentiation(P0.05). MMP-7 expression was significantly correlated with tumor infiltrating degree,Duke's stages and lymph node metastasis(P0.05),but not with age,sex,tumor location,tumor differentiation and distant metastasis(P0.05). Conclusions CDK8 is an oncogene, which can up-regulate MMP-7expression and promote tumor infiltration, invading and metastasis. High expressions of CDK8 and MMP-7indicate that the colorectal cancer patients have a poor prognosis.
