Abstract:Objective To explore the role of tumor necrosis factor alpha(TNF-α) in the development of diabetic nephropathy(DN) in diabetes mellitus(DM) rats.Methods Forty-eight Wistar rats were randomly divided i...Objective To explore the role of tumor necrosis factor alpha(TNF-α) in the development of diabetic nephropathy(DN) in diabetes mellitus(DM) rats.Methods Forty-eight Wistar rats were randomly divided into control group and experimental group.The subjects in diabetes mellitus group were injected intraperitoneally with the dose of STZ 50 mg/kg,while the ones in control group were injected intraperitoneally with the same volume of citric acid buffer.At weeks 6,10 and 14,fast blood glucose(FBG),24 h urine albumine excretive rate(UAER) and the pathological changes of kidney were observed,fast blood insulin(FINS) were measured by ELISA,and the pathological changes of kidney were examined using light microscope and electron microscope.The glomerular volume and glomerular basement membrane(GBM) thickness were measured by computerized image processing.TNF-α expression in kidney was determined by immunohistochemistry.Results With the development of DM,there was more urinary albumin excretion(P0.01),more glomerular matrix and thicker GBM in diabetic rats(P0.01).Compared with the control group,plasma TNF-α increased significantly in experimental group(P0.01),and immunohistochemical results showed that the IDF of TNF-α increased significantly in diabetic rats(P0.01).Conclusion TNF-α may play a negative role in the progression of DN.Read More
