Title: Protective effects of Ginkgo biloba extract on apoptosis of pancreatic β-cells in insulin resistance rats induced by high fat diet
Abstract: Objective To investigate the protective effects of Ginkgo biloba extract on the apoptosis of pancreatic β-cell in insulin resistance rats.Methods 40 male Wistar rats were randomly divided into two groups taking either normal chow(NC group)or high fat diet(HF group) respectively.Four weeks later,insulin resistance models were successfully duplicated,HF rats were randomly divided into hyperlipidemia group(HL),rosiglitazone group(HL+RSG,n = 10),Ginkgo biloba extract group(HL + EGb).We observed the apoptosis rate of pancreatic β-cells by TUNEL and the expression of bax and bcl-2.Results Compared with the NC group,the apoptosis rate of pancreatic β-cell stepped up significantly in the HL group(P 0.05),and the bcl-2 protein expression level of pancreatic β-cell decreased,but the bax protein expression level of pancreatic β-cell increased significantly(P 0.05).Compared with the HL group,the apoptosis rate and the bax protein expression level of pancreatic β-cell in group HL + EGb and group HL + RSG decreased,and the bcl-2 protein expression level of pancreatic β-cell increased.Conclusion The effects of Ginkgo biloba extract on protecting the pancreatic β-cells is closely related to the expression levels of apoptosis-related protein.
