Abstract:Objective To explore the expression changes of c-Jun activation domain binding protein 1(JAB1) in mice peritoneal macrophages at early stage of severe burn injury,and to investigate the effects of dia...Objective To explore the expression changes of c-Jun activation domain binding protein 1(JAB1) in mice peritoneal macrophages at early stage of severe burn injury,and to investigate the effects of diazepam-ketamine on the expression changes of JAB1 in peitoneal macrophages of mice with severe burn injury.Methods Healthy adult Kunming mice were randomly divided into 4 groups,namely control group,severe burn injury group,diazepam-ketamine anesthesia group,and anesthesia after severe burn injury group.The burn model,used third degree burn of 20%-30% of the total body surface area(TBSA).Mice in anesthesia after severe burn injury group were anesthetized with diazepam(0.4mg/kg IM) and ketamine(10mg/kg IM) after burn injury.Mice peritoneal macrophages were collected and total protein was extracted.Western blotting assay was used to detect the protein expression level of JAB1 in peritoneal macrophages of every group mice.Results The protein expression level of JAB1 in peritoneal macrophages of mice was markedly declined at 30min after severe burn injury compared with the control group(P0.05),reached the lowest at 6h(P0.01),resumed to normal level 12h after severe burn injury.However,the protein expression level of JAB1 did not change significantly in peritoneal macrophages of mice with diazepam-ketamine anesthesia after severe burn injury.Conclusion Severe burn injury can significantly decrease the protein expression level of JAB1 in peritoneal macrophages of mice.However,the down-regulation of the protein expression level of JAB1 could be prevented by diazepam-ketamine anesthesia after severe burn injury.Read More
