Abstract:Objective To observe the expression changes of glucocorticoid receptor(GR) in the lung tissues,and to investigate the effects of diazepam-ketamine on the expression changes of glucocorticoid receptor ...Objective To observe the expression changes of glucocorticoid receptor(GR) in the lung tissues,and to investigate the effects of diazepam-ketamine on the expression changes of glucocorticoid receptor in lung tissues of mice with severe burn injury.Methods Seventy healthy adult Kunming mice were randomly divided into 3 groups,namely the normal control group,severe burn injury group,and anesthesia after severe burn injury group.In the latter two groups,the burn model,used third degree burn of the 20%-30% of the total body surface area(TBSA).Mice in anesthesia after severe burn injury group were anesthetized with diazepam(0.4mg/kg IM) and ketamine(10mg/kg IM) after burn injury.Western blot was used to detect the expression level of GR in the lung tissues of every group mice.Results The expression level of GR in lung tissues of mice was markedly declined from 30 minutes after severe burn injury,resumed to normal level after the 12th hour.However,the expression level of GR did not change significantly in the lung tissues of mice with diazepam-ketamine anesthesia after severe burn injury.Conclusion Severe burning can significantly decrease the expression level of GR in the lung tissues of mice,however,the down-regulation of the expression level of GR can be prevented by diazepam-ketamine anesthesia after severe burn injury.The results suggest that diazepam-ketamine anesthesia might partially improved the state of stress induced by severe burn injury,which may correlate with inhibition of down-regulation of the expression level of GR.Read More
