Abstract:Objective To observe the effects of liqustazine on expression change of glucose-regulated protein 94(GRP94) in the cardiomyocytes of severe burn rats.Methods Healthy adult Wistar rats were randomly di...Objective To observe the effects of liqustazine on expression change of glucose-regulated protein 94(GRP94) in the cardiomyocytes of severe burn rats.Methods Healthy adult Wistar rats were randomly divided into the control group(without burns),the burn group(subjected to 30%TBSA III degree scalding) and the treatment group(treated with intraperitoneal injection of Liqustazine(20 mg/kg) immediately after scald injury).The myocardial tissue samples were harvested at 1,3,6,12 and 24 hours post burn(PBH).The cardiac troponin T(cTnT) and tumor necrosis factor alpha(TNF-α) contents in plasma were measured by enzyme linked immunosorbent assay method(ELISA) and radioimmuno assay(RIA) respectively,RT-PCR and immunohistochemistry were applied to monitor the gene expression of GRP94.Results the content of cTnT in plasma was significantly increased 3 hours post burn(P0.01),the gene expression of GRP94 and caspase-3 activity 3 hours post burn reached the peak after 12 h,while the GRP94 expression in Liqustazine treatment group was dramatically decreased.Conclusion Severe burn can induce GRP94 expression increase in rat cardiomyocytes,Liqustazine has notable inhibitory effects on GRP94 expression increase in the cardiomyocytes during the early stage of postburn.Read More
