Title: Neuroprotective effect of aspirin preconditioning on cerebral ischemia reperfusion injury in Sprague-Dawley rats
Abstract: Objective To explore the neuroprotective effect of aspirin preconditioning on cerebral ischemia reperfusion injury in Sprague-Dawley(SD)rats and its mechanism. Methods Thirty-six adult healthy male SD rats were randomly divided into sham group,ischemia group,aspirin preconditioning group.The middle cerebral artery occlusion/reperfusion(MCAO/R)model was made by the suture method.After 2 h ischemia and 24 h reperfusion,the animals were neurologically assessed with a 5 point scale,and then beheaded.The volume of cerebral infarction was measured by TTC staining.The expression of caspase 3 was detected by immunohistochemistry,and the numbers of neuron apoptosis were detected by TUNEL. Results Compared with ischemia group,the volume of infarction significantly reduced in aspirin preconditioning group(P0.05),the neurological scores were improved(P0.05),the expression of caspase 3 and the numbers of neuron apoptosis significantly decreased(P0.01). Conclusion The results suggest that neuroprotective mechanism may be related to the decrease of the expression of caspase 3 protein and the numbers of TUNEL positive cells.
