Title: Neuroprotective mechanism of Butylphthalide injection pretreatment on protect cerebral ischemica reperfusion injury viathrough the PI3K/Akt pathway in rats
Abstract: Objective To study the neuroprotectionve mechanism of Butylphthalide injection pretreatment on cerebral ischemia reperfusion injury in rats and explore the possible mechanism effect viaof PI3K/Akt pathway.Methods The healthy adult male SD rats were randomly divided into the sham-operative group,focal cerebral ischemia reperfusion group and butylphthalein preconditioning group.After ischemia for 2 hours and reperfusion for 24h,the neurological scores were taken to evaluated the rats,the infarction volume were measured by TTC staining,the morphological feature of neurons was observed by HE staining,the level of caspase-3 and p-Akt were detected by immunohistochemistry.Results Compared with sham-operation group,there were more obvious neurologic impairment,infarction volume,cell injury and caspase-3,p-Akt positive cells in focal cerebral ischemia reperfusion group.Compared with focal cerebral ischemia reperfusion group,the neurological impairment were alleviated,the infarction volume were reduced,cell injury were relieved,caspase-3 positive cells were reduced and p-Akt positive cells were increased in butylphthalein preconditioning group.Conclusion Butylphthalein could protect neuron through up regulation of p-Akt expression and down regulation of caspase-3 expression.
