Title: Optimizing conditions of chitinase production from Paecilomyces lilacinus strain FZ-0289
Abstract: Single-factor design and multiple factor design were used for optimiz ing cultural conditions of chitinase production from Paecilomyces lilacinus stra in FZ-0289. Single-factor design showed that chitin powder (165 μm) was the b est medium for inducing the chitinase from FZ-0289, and by adding peptone to c hitin powder the peak of chitinase production could appeared ahead of time. Mult iple factor design showed that optimal cultural medium for chitinase production included 2 g·L~-1 chitin powder (165 μm), 2 g·L~-1 peptone, 1 g·L~-1 KH2PO4, 0 .5 g·L~-1 MgSO4, 0.2 g·L~-1 CaCl2 and 0.2 g·L~-1 NaCl. The FZ-0 289 strain was activated on potato sugar agar (PSA) plates for 10 days, then was inoculat ed with 6×10~9 spores·L~- 1 in 150 mL optimal liquid medium. After fermented at 28 ℃ with the revolving speed of 150 r·min~-1 in 500 mL conical-flask, the chitinase activity co uld reach 0.11 U·mL~-1 on the third day of incubation period.
