Title: A comparative study on the application of two types of nucleic acid stain
Abstract:Objective To investigate the staining effect of GoldView to the DNA/RNA samples in the gels and evaluate the possible application of GoldView in routine nucleic acid staining as a substitute for EtBr....Objective To investigate the staining effect of GoldView to the DNA/RNA samples in the gels and evaluate the possible application of GoldView in routine nucleic acid staining as a substitute for EtBr. Methods After the run was completed, the gels were incubated in GoldView(1:20,000 dilution)or EtBr(0.5μg/ml)solution for about 30~45 minutes with gentle rocking. During staining, gels were protected from light. Visualize and photograph the gels by placing it on a 300nm UV transilluminator. Results GoldView-dsDNA complexes emitted green fluorescence and GoldView-ssDNA complexes red one when being exposed to 300 nm UV light. GoldView stain detected as little as 5 ng per band of dsDNA compared to 2.5 ng with EtBr.The two stains performed with approximately equivalent sensitivity for detecting dsDNA.
