Abstract:Objective To compare the sensitivity of ethidium bromide(EB)and three types of new nucleic acid dye:GoldViewna II,GoldView and DNA Green staining DNA by three staining methods:prestaining gel method,p...Objective To compare the sensitivity of ethidium bromide(EB)and three types of new nucleic acid dye:GoldViewna II,GoldView and DNA Green staining DNA by three staining methods:prestaining gel method,prestaining sample method and poststaining method.Methods Four nucleic acid dyes were used to stain DNA respectively by three staining menthods in agarose gel electrophoresis and the results were observed and analyzed.Results The comparative result of nucleic acid dye:the sensitivity of GoldViewna II is highest of all;the sensitivities of DNA Green and EB are in the same degree;the sensitivity of GoldView is less than EB.The comparative result of staining method:The sensitivity of staining DNA by prestaining gel method is highest of the three and poststaining method is higher than prestaining sample method.Conclusion These three types of new nucleic acid dye can substitute for EB as a general dye to stain DNA in agarose gel electrophoresis.It is recommended to use Prestaining gel method when using new type of nucleic acid dye.Read More
