Title: Inhibition of hyperbaric oxygen on iNOS expression of gerbil to alleviate ischemic nueronal apoptosis
Abstract: Objective To investigate the effects of hyperbaric oxygen(HBO) on nitric oxide(NO) synthesization and the protection of neurons in gerbils′ cortex following cerebral ischemia/reperfusion.Methods The gerbil models of cerebral ischemia/reperfusion were established by bilateral carotid artery clamping for 30 min. The nitric oxide was detected by using electrical chemistry method, the expressions of nitric oxide synthase(NOS) and the apoptotic neurons were probed by immunochemistry technique in gerbils′ cortex.Results Following cerebral ischemia/reperfusion,the NO increased very evidently in gerbils′ cortex. The expressions of three isoforms of NOS were detectable in the same areas. Especially in 2nd day, the expression of the inducible NOS was most obvious. Meanwhile the NO increased to maximum too.The apoptotic neurons could be all detected in gerbils′ cortex in 1st,2nd ,3rd day following ischemia/reperfusion, Especially in 2nd and 3rd day, the apoptotic neurons was more obvious. HBO could inhibit the the expression of the inducible NOS, lessen the NO releasion,alleviate the neuronal apoptosis.Conclusion HBO can alleviate the neuronal apoptosis, protect brain by inhibiting the NO synthesization during cerebral ischemia/reperfusion in gerbils′ cortex.
