Title: Hyperbaric Oxygen Inhibits NO Synthesization and Improves Cerebral Ischemic Cell Apoptosis
Abstract: Objective To investigate the effects of hyperbaric oxygen(HBO)on nitric oxide(NO)syntheˉsization and the protection of neurons in gerbils'cortex following cerebral ischemia/reperfusion.Methods The gerˉbil models of cerebral ischemia/reperfusion were established by bilateral carotid artery clamping for30min.Nitric oxide was detected by using electrical chemistry method,expressions of nitric oxide synthase(NOS)and the apopˉtotic neurons were probed by immunochemistry technique in gerbils'cortex.Results Following cerebral ischemiˉa/reperfusion,NO increased very evidently in gerbils'cortex.Expressions of three isoforms of NOS were detectable in the same areas.Especially on2nd day,expression of the inducible NOS was most obvious.Meanwhile NO inˉcreased to maximum too.The apoptotic neurons could be all detected in gerbils'cortex on1st,2nd and3rd day following ischemia/reperfusion,Especially on2nd and3rd day,the apoptotic neurons were more obvious.HBO could inhibit the the expression of the inducible NOS,lessen the NO release,alleviate neuronal apoptoˉsis.Conclusions HBO can alleviate neuronal apoptosis,protect brain by inhibiting the NO synthesization during cerebral ischemia/reperfusion in gerbils'cortex.
