Abstract:Objective To study the relationship of apoptosis-inducing factor(AIF) translocation and dopaminergic neurons damage in substantia nigra of Parkinson' s disease(PD) model mice.Methods 1-methyl-4-phenyl...Objective To study the relationship of apoptosis-inducing factor(AIF) translocation and dopaminergic neurons damage in substantia nigra of Parkinson' s disease(PD) model mice.Methods 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced PD model was performed in mice.The midbrains were removed at 2h,24h and 72h time points to observe the destruction of dopaminergic neurons by tyrosine hydroxylase(TH) immunohistochemical analysis.AIF translocation was observed by immunohistochemistry and Western blot.Results Compare with control group,the number of TH positive neurons reduced with time prolongation.Mitochondrio-nuclear translocation of AIF was found in substantia nigra of PD group,reached peak in number at 2h,then reduced gradually with time prolongation in PD group.Conclusions AIF translocation is an early marker of dopaminergic neurons damage in substantia nigra of PD mice and may play an important role in PD pathogenesis.Read More
