M...Objective To investigate biological effect of paraquat (PQ),environmental factor,on dopaminergic neurons of substantia nigra (SN),and the role of matrix metalloproteinases (MMP) in the pathogenesis. Methods The C57BL mice were received intraperitoneal injections of 10 mg/kg PQ,and HPLC was used to detect the content of dopamine (DA) in striatum. Immunohistochemistry assay was used to measure the immunoreactivity of tyrosine hydroxylase (TH) in SN. Results Compared with that in control group,the amount of TH positive neuron in MPTP group was significantly decreased(P0.01),and the content of DA in striatum was decreased(P0.01),the expressions of MMP-2 and MMP-9 were significantly increased(P0.01) after treated with PQ.Conclusions PQ might injury dopaminergic neurons of substantia nigra,MMP-2 and MMP-9 might attend the pathogenesis in C57BL mice.Read More
