Title: The experimental study of rAd-p53 in treatment of rabbits VX2 brain tumors guided by MRI
Abstract: Objective:To evaluate the feasibility of MRI-guided injection rAd-p53 to the VX2 brain tumor of rabbit.Methods:12 New Zealand white rabbits were randomly divided into control group(group A),treatment group(group B), with 6 rabbits in each group.Under general anesthesia through the skull hole of planting methods,VX2 transplanted brain tumor model was established.The rabbits in group A and group B were implanted VX2 brain tumor respectively.When tumors about 0.5 cm in diameter,the MRI-guided injection of 0.5 ml sterile saline and 0.5 ml rAd-p53 were performed respectively.Subsequently,group A,B after 1,3,5 days were examined by MRI,then two rabbits in each group were killed and pathological examination and immunofluorescence were performed.Results:Brain tumor grew in aec 12 rabbits.The tumors showed slight low signal on T1WI and slight high signal on T2WI as well as contrast enhancement after injection of Gd-DTPA.The maximum diameter of brain tumor was from 0.5 to 0.7 cm.Over the same period.MRI showed a consistent tumor size and shape.MRI findings were in line with pathological changes.It could meet the needs of interventional therapy.The procedures of sterile saline and rAd-p53 injection were all performed successfully.Pathology and MRI manifestation had no significant differences after postoperative 1,3,5 days.But in immunofluorescence,P53 protein expression and apoptosis in group B were significantly saperior to group A.Conclusion:It is safe and feasibility of rAd-p53 injection into brain tumor under MRI-guidance,which could significantly increase the P53 protein expression and tumor apoptosis.
