Title: Expression of MAD2 in colorectal cancer and its clinical significance.
Abstract: Objective To study the expression of gene MAD2 in colorectal cancer and the relation between the expression of gene MAD2 and colorectal cancer,adenoma and normal tissue and to evaluate their clinical significances. Methods Immunohistochemistry method and real-time fluorescence quantitative PCR were used to analyze the expression of gene MAD2 in the colorectal cancer,adenoma and normal tissue.PCR amplification and DNA sequencing were performed to detect the mutations of gene MAD2. Results The expression of MAD2 in colorectal cancer was obviously higher than that in adenoma and normal tissue by inmmunohistochemistry method(66.7% vs 39.6% vs 22.9%). There was significant difference among colorectal cancers,adenoma and normal tissue (P0.001),and it was related to tumor differentiation and survival time (P0.05). The quantity of MAD2 in colorectal cancer was obviously higher than that in normal tissue by real-time fluorescence quantitative PCR and no mutation of gene MAD2 was found in colorectal cancer. Conclusion Positive expression of MAD2 is mainly involved in colorectal carcinogenesis and MAD2 may be associated with prognosis of colorectal cancer.
