Title: Interleukin-27 affects serum IFN-gamma and IL-33 activities in patients with bronchial asthma
Abstract: Objective To study the effect of interleukin (IL)-27 on INF-γ and IL-33 activities in patients with bronchial asthma (asthma).Methods Two groups of experiments,the serum levels of IFN-γ and IL-27 and IL-33 in asthmatic patients or control group by ELISA.The levels of WBC and neutrophils and EOS by hematology analyzer.PBMCs from 33 asthmatic patients were cultured with recombinant human IL-27 (5 μg/L or 0.5 μμg/L) for 12 hours in vitro,and then the levels of IFN-γ and IL-33 in their supematant were measured by ELISA.The explore of correlation between IL-27 activating factor and IL-33,IFN-γ in the asthmatic patients.Results The levels of IL-33 in asthmatic patients were significantly increased and IFN-γ,IL-27 had a significant decrease compared with that in healthy control group (t IFN-γ =27.52,P IFN γ < 0.01 ; t IL-27 =32.77,P IL-27 < 0.01).The values of IFN-γ in supematant were (85.9± 12.1) μg/L,(19.6±3.2) μg/L compared with that in cytokine stimulated control group (11.3±2.7) μg/L had increased significantly (t =34.57,P <0.01; t =11.39,P <0.01).The group treated with 5 μg/L IL-27 had decreased significantly IL-33 levels compared with the cytokine stimulated control group [(33.2±11.8) μg/L vs (116.9±17.6) μg/L,t =22.69,P <0.01].There was a positive correlation between IFN-γ and IL 27 in asthmatic patients.Conclusions The levels of IL-27,IFN-γ,IL-33 were interelated.
