Title: Effect of Polyphylin I on Proliferation and VEGF Expression of Ha Ca T Cells under Hypoxia
Abstract: Objective To study the effects of Polyphyllin Ⅰ on proliferation and VEGF expression of HaCaT cells mediated by hypoxia. Methods MTT assay was performed to investigate the effects of Polyphyllin Ⅰ on the growth of HaCaT keratinocytes under normoxic and hypoxic conditions. ELISA was used to detect VEGF protein secreted by the cells under normoxic and hypoxic conditions. Real-time quantitative reverse transcriptase polymerase chain reaction was used to detect VEGF mRNA expression and Western blotting to detect VEGF,HIF-1α,STAT-3,phospho-STAT-3 expression. Results Within the selected concentration( 0. 75,1. 5,3,5μg / m L),Polyphyllin Ⅰ inhibited HaCaT keratinocytes cell proliferation and induced a significant reduction in VEGF mRNA under hypoxia conditions. Polyphyllin Ⅰ treatment inhibited hypoxia-induced expression HIF-1α,STAT-3,phospho-STAT-3,VEGF under normoxic and hypoxic conditions. Conclusion PolyphyllinⅠ downregulates VEGF expression in HaCaT keratinocytes by inhibiting the activity of STAT3 and HIF-1 α
