Title: Relationship among Tumor Microvessel Density, nm23 - H1 expression, PCNA index and Prognosis in Patients with Osteosarcoma
Abstract: Objective To investigate relationship among the tumor microvessel density (MVD) ,nm23-H1 expression, proliferating cell nuclear antigen (PCNA) index and the prognosis in the patients with osteosarcomas. Methods MVD, nm23-H1 expression, and PCNA index were studied by immunohistochemical technique in 80 cases of osteosarcomas. The prognostic factors were also analyzed with cox multivariate model. Results The MVD was positively correlated with PCNA index,and negatively with nm23-H1 expression. Cox multivariate model analysis confirmed that the MVD,nm23-H1,and PCNA were important prognostic factor.The prognosis was better in the patients with low MVD than that in the patients with high MVD ( P 0.05) . In those patients with high MVD, high expression of nm23 - H1 allowed the identification of a subgroup with significantly higher survival rate ( P 0.05). Conclusions The prognosis and metastasis of osteosarcoma depends very much on its in situ microenvironment. But the high MVD appears necessary but not sufficient for the metastasis to occur. The MVD may interact with nm23-H1 and PCNA, and their interaction contributes to the metastasis and prognosis of osteosarcoma.
