Title: Quantitative Expression of hTERT mRNA in NSCLS and Its Relation with COX-2 Gene Expression
Abstract: Objective: To investigate the expression of hTERT mRNA in human non small-cell lung cancer(NSCLC) and its relations with that of COX-2 gene,and to discuss the possible modulation mechanism of activating hTERT.Methods: The hTERT mRNA expression was detected by quantitative reverse transcription-polymerase chain reaction(RT-qPCR).The COX-2 gene protein expression was detected by immunohistochemistry.The correlation analysis was used to estimate their relation.Results: The hTERT mRNA level in NSCLC tissue from 45 patients was higher than in normal lung tissue from 21 normal controls(2.854±0.728 vs 2.042±0.378,P0.01).There were no significant associations between NSCLC hTERT mRNA expression level and patient's age,gender,degree of differentiation,lymph node metastasis and clinical stages(all P0.05),but the hTERT mRNA expression level in non-small cell lung cancer was higher than in normal tissue(P0.05).Correlation analysis revealed that expression of COX-2 was significantly related to that of hTERT(r=0.495,P0.01).Conclusion: hTERT may be useful in the clinical diagnosis of NSCLC.The significant correlation between the expression of hTERT and COX-2 indicates that one of the approaches of hTERT activation and up-regulation may be conferred by overexpression of COX-2.
