Title: Expression law of VEGF-D and VEGFR-3 and the relationship between VEGF-D/VEGFR-3 and lymphatic metastasis in thyroid parpillary carcinoma
Abstract: OBJECTIVE:To investigate the effect of VEGF-D and VEGFR-3 on the process of thyroid papillary carcinoma lymphatic metastasis. METHODS: Expression levels of VEGF-D and VEGFR-3 in carcinoma,para-carcinoma and normal tissue of 45 thyroid parpillary carcinoma patients (15N0 cases, 30N+ cases) were detected with the reverse transcription polymerase chain reaction (RT-PCR), and the law of them was explored. RESULTS: In lymph metastasis group, the expressions of VEGF-D and VEGFR-3 were 1.602±0.070,1.500±0.060 in carcinoma area, 1.139±0.064,1.156±0.060 in para-carcinoma area, 0.444±0.046, 0.447±0.059 in normal areas respectively. In non-lymph metastasis group, the levels of VEGF-D and VEGFR-3 were 1.304±0.071,1.307±0.053 in carcinoma area, 0.898±0.070, 0.882±0.101 in para-carcinoma area and 0.375±0.032,0.318±0.057 in normal areas respectively. The expression law of VEGF-D and VEGFR-3 was that the expression in carcinoma tissue was higher than those in para-carcinoma (t≥4.03, P≤0.001), and normal tissue (t≥5.486,P=0.000), and the difference was significant; the expression in carcinoma and para-carcinoma of N+ group was higher than that of N_0 group, t≥2.339,P≤0.028, the difference was significant; the difference between the expression in normal tissue of the 2 groups was insignificant, t≤1.572, P≥0.124. CONCLUSIONS:The expressions of VEGF-D and VEGFR-3 in thyroid papillary carcinoma are correlated with lymphatic metastasis. Their high expressions in carcinoma and para-carcinoma may predict lymphatic metastasis. The pathway may become a new target for preventing and treating thyroid papillary carcinoma lymphatic metastasis.
