Title: Effect of celecoxib on endometrial carcinoma cell proliferation and expression of COX-2 mRNA
Abstract: Objective To investigate the effect of celecoxib on endometrial carcinoma cell HEC-1-B and expression of the COX-2 mRNA.Methods To investigate the inhibition of different concentration Cele- coxib on the growth of human endometrial carcinoma cell HEC-1-B by the method of cell culture and MTT experiments.Detect the changes of expression of the COX-2 mRNA by RT-PCR.Results Celecoxib could effectively inhibit the growth of the HEC-1-B cell in dose and time dependent manner.Celecoxib could inhib- it the expression of the COX-2 mRNA.The inhibition ratio of expression in COX-2 mRNA of 20μmol/L and 50μmol/L Celecoxib is 25.92 % and 50.81% respectively. Conclusion Celecoxib may inhibit the growth of endometrial carcinoma cell by degrading the expression of the COX-2 mRNA.
