Title: Celecoxib suppressed COX-2 protein expression and induced apoptosisin human breast cancer cell line MDA-MB-231
Abstract: Objective To study the effects of celecoxib in inducing apoptosis and suppressing the expression of cyclooxygenase-2 protein in human breast cancer cell lines MDA-MB-231.Methods The human breast cancer cell line MDA-MB-231 was studied and SGC-7901 served as the positive control cell line.Cells were cultured for 24 h with celecoxib at different concentrations.Apoptosis was detected by DNA fragmentation electrophoresis,while the COX-2 protein by Western blot.Image Tool software was used to analyze the Western blotting image.Results With the increase of celecoxib concentrations,cell apoptosis was increased.The COX-2 protein expression could be obviously suppressed by COX-2 specific inhibitor celecoxib.The concentration of maximum suppressing effect was 100 μmol/L.Softwareanalysis revealed that with the increase of celecoxib concentrations,the expression ofCOX-2 protein was obviously decreased with the difference being significant (P0.01).Conclusion Celecoxib could effectively induce apoptosis in MDA-MB-231 cell line,which may be related to the expression inhibition of COX-2 protein.
