Title: Protective effect on kidney and mechanism of Chinese medicinal with actions of replenishing qi,nourishing yin,removing accumulation and free collateral vessels in rats with early diabetes
Abstract: Objective To investigate the protective effect of Chinese medicinal with the actions of replenishing qi,nourishing yin,removing accumulation and free collateral vessels on kidney in rats with early diabetes,and discuss possible molecular biology mechanism.Methods SD rats were randomly divided into the nephrectomy group,model group,irbesartan group,high-dose,mid-dose and low dose Chinese medicinal groups(high-dose group,mid-dose group and low-dose group).All groups were given relevant medicinal intragastrically.At the end of the 6th week the levels of 24-hour urinary protein(U Pro) and renal function were detected in rats of all groups.The pathological changes of kidney were observed with light microscope.The protein expressions of p38MAPK,p-p38MAPK,STAT3 and p-STAT3 in renal cortex were detected by using Western blotting method.Results The level of U Pro and protein expressions of p-p38MAPK and p-STAT3 decreased significantly in the irbesatan group,low-dose group and mid-dose group compared with the model group(P0.05).The renal function was improved and the pathological changes were relieved in all treatment groups compared with the model group(P0.05).Conclusion The inhibition of activation of p38MAPK/STAT3 signal transmission pathway may be one of kidney-protecting mechanisms of Chinese medicinal with the actions of replenishing qi,nourishing yin,removing accumulation and free collateral vessels in rats with early diabetes.
