Abstract:Objective To study the influence and discuss the mechanism of RNA interference Caspase-3 gene on apoptosis of cardiomyocyte apoptosis induced by hypoxia.Methods Hypoxia cardiomyocytes were treated wit...Objective To study the influence and discuss the mechanism of RNA interference Caspase-3 gene on apoptosis of cardiomyocyte apoptosis induced by hypoxia.Methods Hypoxia cardiomyocytes were treated with siRNA targeted for caspase-3 gene.During different time span,the treated cardiomyocytes were detected for the occurrence of myocardial apoptosis,LDH leakage,calcium concentration,MDA content,SOD activity.In addition,the expression of caspase-3 gene and its protein in cardiomyocytes were assayed by the methods of reverse transcription polymerase chain reaction(RT-PCR)and Western-blot analysis respectively.Results The occurrence of myocardial apoptosis was increased with the time of hypoxia.LDH leakage,calcium concentration,MDA content were increased,SOD activity was declined in hypoxia cardiomyocytes.The expression of Caspase-3 mRNA and protein in cardiomyocytes induced by hypoxia for RNA interference group significantly decreased when compared with controls and scrambled sequence group.Conclusion The occurrence of apoptosis in hypoxia cardiomyocytes was significantly inhibited by RNA interference Caspase-3 gene.It was suggested that Caspase-3 gene could play a central role in apoptosis of hypoxia cardiomyocytes.Read More
