Abstract:Objective To investigate the effects of Tangkang on rats with diabetic retinopathy and further elucidate the mechanism,in order to provide the experimental basis for Tangkang preventing and curing dia...Objective To investigate the effects of Tangkang on rats with diabetic retinopathy and further elucidate the mechanism,in order to provide the experimental basis for Tangkang preventing and curing diabetic retinopathy.Methods The normal Wistar rats were given streptozotocin(STZ)in intraperitoneal injection to induce the diabetic rats model.75 rats were divided into 6 groups,including normal group,modeling group,three Tangkang treatment groups(high,median and low dosage)and Danakang group.The body weight and non fasting blood glucose of all experimental groups were tested every week.After 12 weeks,retinal vascular preparations were performed by trypsin digestion to observe the histopathologic alternations of retinal microvascular;Western blot were used to detect the protein expression of NF-κBP65,the mRNA expression of Bax and IL-1β in retina of all experimental groups were detected by reverse transcription polymerase chain reaction(RT-PCR).Results Compared with the normal group,the numbers of acllular capillaries were significantly decreased(P0.01),in all treated groups,and the high dosage of group was the most significant.The results of Western blot showed that the protein expression of NF-κBP65 in retinal tissue of model group was increased markly compared with age-matched normal control group(P0.01).All treatment groups showed that the protein expression of NF-κBP65 was inhibited.Compared with age-matched normal control group,the mRNA expression of IL-1β and Bax were increased in the retina of rats of modeling group(P0.01).All treatment groups showed that the mRNA expression of IL-1β and Bax were inhibited.Conclusion Tangkang could inhibit the protein expression of NF-κBP65 and mRNA expression of IL-1β and Bax,and the development of histopathology in the retina of diabetic rats,showed protective role to the microcapillary in the retina of diabetic rats.Read More
