Abstract:In order to study the protection of OmpH and OmpA proteins of Pasteurellosis multocida derived from yak,the prokaryotic expression plasmid pET28a-OmpH and pET28a-OmpA were constructed.The vaccines wer...In order to study the protection of OmpH and OmpA proteins of Pasteurellosis multocida derived from yak,the prokaryotic expression plasmid pET28a-OmpH and pET28a-OmpA were constructed.The vaccines were prepared with the purified recombinant outer member protein H(rOmpH) and the recombinant outer member protein A(rOmpA).The mice were hypodermicly immunized with rOmpH,rOmpA,rOmpH-A vaccines and the inactivated whole bacteria vaccine,respectively and PBS was used as negative control.Sera were collected each week from the mice to examine antibodies after first immunization using an indirect ELISA.The immunized mice were challenged with 2LD50 yak P.multocida on day 9 after third immunization.In result,the protection rate of the inactivated whole bacteria vaccine and rOmpA were 80% and 40% respectively,but mice in the other groups were dead.The rOmpH and rOmpH-A groups only postponed the time of mouse death,and the protectivity was weak and the immunization was unable to resist the lethal attack using homologous strain.The results showed that the rOmpA protein had protective efficacy,and the rOmpH had not protective efficacy.Read More
