Abstract:A high performance liquid chromatography(HPLC) method was developed to determine chlorogenic acid,baicalin,quercetin,luteolin and chrysin in Chrysanthemum samples.The HPLC conditions were as follows: ...A high performance liquid chromatography(HPLC) method was developed to determine chlorogenic acid,baicalin,quercetin,luteolin and chrysin in Chrysanthemum samples.The HPLC conditions were as follows: Nov-pak C18(250×3.9 mm i.d.,5 μm) column,0.1% phosphoric acid aqueous solution-methanol as mobile phase with gradient elution,flow rate with 0.6 mL/min,254 nm as the optimum detection wavelength.Good linear relationship was established within 10-3~10-5 g/mL.RSDs of retention time and peak area of the five standard samples ranged from 0.20% to 0.96%,and from 0.23% to 0.77%,respectively,showing excellent reproducibility.The recoveries were between 96.5% and 104.0%,and the determination result was accuracy.Additionally,the fingerprints of Chrysanthemum were established with the included angle cosine and correlation coefficient methods to calculate the similarity of 12 Chrysanthemum samples.According to the result of correlation coefficient,the MATLAB procedure was applied for the cluster analysis,and the result is consistent with both the genera and the species of the actual samples.It provides the criterion for quality control and discrimination of Chrysanthemum.Read More
