Abstract:A precise and sensitive method was developed for the simultaneous determination of ten polyphenolic compounds in tea leaves by high performance liquid chromatography(HPLC).The ten compounds were extra...A precise and sensitive method was developed for the simultaneous determination of ten polyphenolic compounds in tea leaves by high performance liquid chromatography(HPLC).The ten compounds were extracted by 20% ethanol solution from the tea powder,and separated on an Elite-SinoChrom ODS-BP column(5 μm,250 nm × 4.6 mm i.d.) at 30 ℃.A gradient program was carried out at a flow rate of 1.0 mL/min using methanol and 1% acetic acid solution as the eluent.The diode assay detection was used for qualitative and quantitative analysis and the detection wavelength was set at 280 nm.The results indicated that the chromatographic peaks of 10 compounds and their corresponding spectra were ideal under the optimal conditions.The calibration curves of ten analytes were linear in the range of 0.16 ~ 1740 mg/L,with correlation coefficients more than 0.998.The limits of detection(LODs) of ten analytes were in the range of 0.82 ~ 48.50 ng(S/N = 3).The method could provide reference for evaluating the quality of tea and other related products.Read More
