Title: Determination of emodin, caffeic acid, chlorogenic acid, scutellarin, luteolin, and polydatin in Reyanning Granules by UPLC
Abstract: Objective To develop a UPLC method for simultaneous determination of emodin, caffeic acid, chlorogenic acid, scutellarin, luteolin, and polydatin in Reyanning Granules. Methods UPLC chromatography was used. The chromatographic separation was carried out on an Acquity UPLC HSS C18 column(100 mm × 2.1 mm, 1.7 μm) with [methanol- acetonitrile(21∶79)]- 0.1% phosphoric acid as mobile phases at the flow rate of 0.5 m L/min for gradient elution. Detection with variable wavelength was used, and the column temperature was 35 ℃ with injection volume of 1.0 μL. Results There were good separations for six components in the specific ranges(r≥0.999 3). The average recovery was in the range of 99.08% — 102.31% with RSD value less than 2.53%(n = 6). Conclusion Simultaneous determination method of emodin, caffeic acid, chlorogenic acid, scutellarin, luteolin, and polydatin in Reyanning Granules by UPLC is established, and is accurate, sensitive, credible, and repeatable which can be applied to the quality control for Reyanning Granules.
