Title: Effect of arsenic trioxide on proliferation and apoptosis of MDA-MB-231 cells
Abstract: Objective To observe different concentrations of arsenic trioxide on the proliferation and apoptosis of the triple negative breast cancer MDA-MB-231 cells.Methods The growth inhibition of different concentrations(0.2 0.4,0.6,0.8,1.0 μmol·L-1)of arsenic trioxide on triple negative breast cancer MDA-MB-231 cells for 24,48,72 h was detected by means of MTT assay.The protein expression of PCNA was detected by Western blotting.Results When the MDA-MB-231 cells were treated with different concentrations of arsenic trioxide for 24 h,48 h,72 h,with the increase of time and concentration,cell proliferation inhibition rate both increased,meanwhile 1.0 μmol·L-1 was most obvious.besides,western blot detection showed that,compared with control group,the expression of PCNA protein decreased with the increase of the concentration and 1.0 μmol·L-1 was most obvious as well.Conclusion Arsenic trioxide can promote apoptosis of MDA-MB-231 cells,which may be related with the decrease of PCNA expression in pro-apoptotic pathway.
