Abstract:Objective To analyze the expression of E-cadherin,Cyclin D1 and β-catenin in the tissues of gastric carcinoma and its lymphatic metastasis,and to explore their relationship and the role in tumor cell ...Objective To analyze the expression of E-cadherin,Cyclin D1 and β-catenin in the tissues of gastric carcinoma and its lymphatic metastasis,and to explore their relationship and the role in tumor cell differentiation,invasion and metastasis.Methods The expression of E-cadherin,Cyclin D1 and β-catenin was detected by immunohistochemistry (IHC) in 50 cases of gastric carcinoma and gastric specimens from 20 patients with dysplasia and 10 healthy controls.Results Membranous staining was observed in control specimens for all components of the complex.Abnormal expression rate of E-cadherin,Cyclin D1 and β-catenin in gastric carcinoma (84%,72%,78%) was significantly higher than that in gastric mucosal hyperplasia (50%,60%,60% respectively,P0.05).The decreases in E-cadherin,Cyclin D1 and β-catenin tumors occurred more frequently in undifferentiated gastric carcinoma (P0.05).There was higher abnormal expression rate of E-cadherin in the patients with tumor infiltrating out of serosa and with lymph node metastasis (P0.01).Up to 60% of gastric dysplasials and 90% of tumors stained abnormally for one or more components of the E-cadherin complex was significantly higher than for no one components of the E-cadherin complex (P0.05).Conclusion E-cadherin and β-catenin expression is associated with invasion and metastasis of gastric carcinoma.The abnormal expression of β-catenin activates the expression of Cyclin D1,and results in unchecked proliferation and differentiation of the cell.E-cadherin complex plays a critical role in the course of change from gastric mucosal hyperplasia to gastric carcinoma.Thus,study of all the components of E-cadherin,Cyclin D1 and β-catenin complex may be more valuable than single component of the complex for the detection of patients with gastric carcinoma.And these are important factors for judging invasion of gastric carcinoma.Read More
