Title: Expression and significance of HIF-1α,GLUT-1 and MMP-2 in patients with non-small cell lung cancer
Abstract: Objective To investigate the expression and clinical significance of HIF-1α, GLUT-1 and MMP-2 in human non-small cell lung cancer(NSCLC) tissues. Methods The expression of HIF-1α, GLUT-1 and MMP-2 was detected using immunohistochemical S-P method and reverse transcription-polymerase chain reaction analysis(RT-PCR) in 80 specimens of NSCLC and 26 specimens corresponding adjacent tissues.Their interrelations and relationship with the pathological characteristics of the tumors were analyzed.Results In NSCLC,the positive expression rates of HIF-1α, GLUT-1 and MMP-2 mRNA were higher than those in normal tissues(P0.01).The expressions of them were higher in the patients with lymph node metastasis or in Ⅲ-Ⅳ stage than those without lymph node metastasis or in Ⅰ-Ⅱ stage(P0.05 or P0.01).HIF-1α expression level was positively correlated with GLUT-1 and MMP-2(P0.01).Conclusion The overexpression of HIF-1α, GLUT-1 and MMP-2 closely correlates with progression,invasion and metastasis of lung carcinoma. Combined detection of HIF-1α, GLUT-1 and MMP-2 may be helpful in evaluating the metastasis,staging lung carcinoma,and predicting the prognosis of the patients with NSCLC.
