Title: Separation and purification of total flavonoids from Baihuasheshecao (Herba Hedyotis Diffusa) with macroporous resin
Abstract: Objective To select the best technique and parameter of the separation and purification of the total flavonoids from Baihuasheshecao ( Herba Hedyotis Diffusa) with macroporous resin. Method The types of macroporous resins,concentration of sample solution,pH values,volume of sample,and concentration, quantity and flow velocity of eluant were reviewed by using the single factor investigation taking the content and recovery of the total flavonoids from Baihuasheshecao as the investigative indexes. Result The parameter of the best purification technique was as follows: 0. 2 g/mL sample solution was prepared according to the radio of sample volume ( 0.4 g/mL,crude medicinal volume to resin volume) ,and pH value was 5. 0. The sample solution was rested for one hour in the chromatographic column of AB-8,eluted with deionized water of 8BV at a flow velocity of 3BV/h,and then eluted again with 70% alcohol of 5BV at a flow velocity of 2BV/ h. The content of the total flavonoids was 38. 9% in the obrained fine product,and the transfer rate was as high as 80%. Conclusion AB-8 macroporous resin has a better purifying effect on the total flavonoids from Baihuasheshecao,which is suitable for large scale industrial production.
