Title: Study on Purification Technology of Total Flavonoids from Fructus Auranti by Macroporous Resin
Abstract: Objective To explore the technology of total flavonoids from fructus auranti purified by macroporous resin. Methods The content of total flavonoids from fructus auranti and recovery rate were chosed as indexes, and the static and dynamic adsorption experiments were adopted to screen the optimum technology of total flavonoids from fructus auranti purified by macroporous resin. Results The best macroporous resin was AB-8. The optimal conditions were as follows: the crude drug whose total flavonoids concentration was 0.2 g·mL-1was diluted with water(1:3) until the Ph value of physic liquor was 3-4. The ratio of drug to resin in the sample was 3:1, and the adsorption rate was 3 BV·h-1. The sample received elution of 4 BV water-4 BV 90% alcohol with rate of 2 BV/h, then 90% alcohol eluent was collected. The recovery rate of total flavonoids from fructus auranti purified by AB-8 macroporous resin was more than 95%,dry extract yield was 4.8%. The purity could reach 33.07% before purification, and reach 66.81% after purification. Conclusion The combination property of AB-8 macroporous resin is the best, and can be used for the purification of total flavonoids from fructus auranti.
