Title: Screening Identification of a Chitinolytic Bacterium Strain DL-06 and its Optimal Chitinase Fermentation Conditions
Abstract: Using colloidal chitin as the sole carbon source, a marine bacteria with high productivity of cold-adapted chitinase was obtained from samples collected in Bohai bay in Dalian city of Liaoning Province, China, named as DL-06. The optimal fermentation conditions of cold-adapted chitinase-producing bacteria by single-factor experiments were determined as followed: colloidal chitin 5.0g/L, peptone 5.0 g/L, fermentation temperature 20℃, pH 7.0, inoculums dose 2%, shaking speed 130r/min, aged seawater 1.0 L and fermentation 30 h, respectively. Under these conditions, the maximum activity of cold-adapted chitinase was 6.87 U/ml.
