Title: Protective effect of antithrombotic traditional Chinese medicine on oxidative injury in vitro: flow cytometry and ELISA analysis
Abstract: Objective To investigate the protective effect of antithrombotic traditional Chinese medicine on human umbilical vein endothelial cell (HUVEC) injury induced by hydrogen peroxide (H2O2).Methods Normal control group,oxidative damage model group and Xueshuan Xinmaining tablet treatment group were chosen in our study.The oxidative injured HUVEC models were established by using H2O2.Intervenes before treatment were made by adding antithrombotic traditional Chinese medicine Xueshuan Xinmaining tablets.Cell viability and cell growth cycle changes were observed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) staining and flow cytometry (FCM),respectively.ELISA was employed to detect the contents of Nitric oxide (NO) and nitric oxide synthase (NOS),as well as superoxide dismutase (SOD) level.Results MTT results showed that the cell activity of HUVECs in the oxidative damage model group was significantly decreased as compared with that in the control group; cell growth of the oxidative damage cells was blocked in the G0-G1 period detected by FCM; and ELISA showed that the levels of NO and NOS were decreased significantly and the activity of SOD was decreased in the oxidative damage model group as compared with those in the normal control group (P<0.05); the level of NO was decreased from 31.95 μ mol/L to 24.82 μmol/L and the level of NOS was decreased from 3.13 μmol/L to 2.58 μmol/L; the activity of SOD was decreased from 128.2 U/mL to 109.17 U/mL; these results showed that HUVEC oxidative damage was obvious.Xueshuan Xinmaining tablet treatment could resist the cell injure induced by H2O2 without obvious cell block at G0-G1 periods,and NO and NOS levels were increased significantly and the activity of SOD was increased as compared with those in the oxidative damage model group (P<0.05).Conclusion FCM and ELISA results show that antithrombotic traditional Chinese medicine has obvious protective effect on HUVEC oxidative injury.
