Title: Comparison of antioxidant activities to CHO cell between Daidzein and Genistein
Abstract: Objective To compare the protective effects of Daidzein and Genistein against H2O2 induced oxidative damage in CHO cell line of healthy subjects and to investigate the antioxidant activity of isoflavones. Methods CHO cells injury model was made with 100μmol/L H2O2 treat- ing of 3h, and the treatment group was processed with Daidzein or Genistein of the same dose(0.25~20μmol/L)for 24h. The cellular morphology was examined with microscopy,the cells proliferation was measured by MTT assays and Superoxide dismutase (SOD) activity, and the cell membrane injury was measured by LDH assays, determined lipid peroxidation evaluating malond- ialdehyde (MDA) production. Results The CHO cellular morphology was altered with H2O2 trea- ting for 3h,MTT value decreased and production of MDA and LDH leakage increased obviously what was diferent compared with control group(P0.01).In the group pretreated with Daidizein or Genistein,the cellular morphology recovered,MTT value was increased,the production of MDA and LDH leakage decreased,significantly different compared with H2O2 group(P0.01). Conclusion Daidizein is just as effective as Genistein has strong protective function against oxidative damage in CHO cells.
