Title: Detection of hepatitis B virus genotypes by amplification refractory mutation system
Abstract: Objective To Establish amplification refractory mutation system (ARMS) for hepatitis B virus genotyping assay and detect HBV genotype in patients with chronic hepatitis B by it.Method Based on HBV genome sequence alignment,we designed specific primers and probe,established ARMS for HBV B and C genotyping and detected 50 clinical samples with chronic HBV infection.Results Among 50 samples,genotype B and C which were detected by ARMS assay is 26% (14/50) and 74% (37/50) separately.The results were completely consistent with sequencing results.Genotype C was more prevalent than genotype B in Changzhou area,and 62% patients with genotype C carried A1762T/G1764A double mutation,much higher than genotype B.Conclusion ARMS was a fast and accurate assay for HBV genotype detection.Dominant HBV genotypes in Changzhou area were genotypes B and C,and genotype C carried more A1762T/G1764A double mutation,maybe associated with more sever liver disease.
