Title: Effects of resveratrol on expression of iNOS mRNA and HO-1 mRNA of 6-OHDA induced chronic Parkinson's disease rats
Abstract: AIM To observe whether resveratrol can have a protective effect on 6-OHDA induced chronic Parkinson's disease(PD)rats through inhibiting the experssion of iNOS mRNA and HO-1 mRNA.METHODS Sixty SD rats without rotational behavior were screened,and 20 of them were randomly divided into sham group and normal administration group(resveratrol 20 mg·kg-1·d-1).The PD model rats were induced by injecting 6-OHDA into the right striatum.Fourty valid PD rats were randomly divided into model control group and resveratrol groups(10,20 and 40 mg·kg-1·d-1).All groups were given orally once daily for 10 wk.The neuron ultrastructure of substantia nigra tissues was observed by electronic microscope,the levels of iNOS mRNA and HO-1 mRNA was determined by real-time PCR.RESULTS Resveratrol could alleviate 6-OHDA induced degeneration of substantia nigra dopaminergic neurons in rat.Real-time PCR showed that resveratrol could decrease the level of iNOS mRNA and HO-1 mRNA of substantia nigra in rats(vs.model group,P 0.01).CONCLUSION Resveratrol have a protective effect on 6-OHDA induced chronic PD model rats,the mechanism may be related with the inhibition of the expression of iNOS mRNA and HO-1 mRNA.
