Title: Determination of daidzein, genistein, formononetin and biochanin A in Trifolium pratense L. by HPLC
Abstract: AIM: To establish an HPLC method for the determination of daidzein, genistein, formononetin and biochanin A in Trifolium pratense L. METHODS: A Kromasil C_ 18 column was used with the mobile phase of acetonitrile -0.1%H_3PO_4 (40∶60), the flow rate was 1.0 mL·min -1 the wavelength of detection was set at 254 nm and the column temperature was at 25°C. RESULTS: The linear range of daidzein, genistein, formononetin and biochanin A was 0.0662~0.6620 μg,0.0978~0.9780 μg, 0.0866~0.8660 μg and 0.0992~0.9920 μg, respectively. The average recovery was 98.9% (RSD=0.46%), 98.8% (RSD=1.21%), 98.8% (RSD= 0.71% ) and 97.5% (RSD=1.33%), respectively. CONCLUSION: The method is simple, rapid and accurate.
