Title: Studies on Callus Induction of Taxus. media and Darkening Inhibition in Callus Subculture
Abstract: The induction condition of T. media var. Hickss branch and leaf were studied. The optimum hormone constitution for callus growth and the darkening in callus subculture were also studied. The results indicated that we could get the high efficiency in callus induction by using the tender branch and leaf because of their strong polarization. 2,4-D was more effective in inducing callus than use NAA. The best medium was MS with addition of 2,4-D (2 mg·L~(-1)), NAA (0.5 mg·L~(-1)) and 6-BA (0.2 mg·L~(-1)). While in subculture, 2,4-D was effective in cell growth than NAA and 6-BA was most effective in growth to the callus. We also found that making the subculture in the twentieth and maintaining enough oxygen between the callus and solid medium could inhibit the darkening of callus in some degree. B_5 medium with the addition of 2,4-D(5 mg·L~(-1)), 6-BA(0.5 mg·L~(-1)) and AC(1 g/L) gave optimum cell growth and the darkening in subculture also could be inhibited someway.
