Title: Effect of Different Explants and Medium Composition on Callus Growth of Cephalotaxus fortunei Hook.f
Abstract: To establish and optimize the technology and method of producing high alkaloid yield callus of Cephalotaxus Hook.f.cultivated tree grown in Dalian district transplanted from Lushan arboretum were used as explant source,and the optimum explant type,basic medium composition and concentration of growth regulators in medium and so on,which were factors affecting on callus induction,were examined in a series order.The juvenile stem segments were the optimum explants because of their earlier and higher rate of induction than that of other explants,medium MS+NAA 3.0 mg/L+KT 0.15 mg/L was confirmed callus induction medium in which callus induction rate reached 91%.In callus subculture medium,lower concentration of NAA(1.0-3.0 mg/L)always increased callus growth,but higher concentration of NAA(8.0 mg/L)reduced callus growth,when NAA concentration was suitable,callus growth on MS displayed lighter browning and faster tissue growth than that on B5.and the induction rate inducted in winter is higher than that in summary.The optimization sequence of obtaining a large quantity and high-alkaloid yield callus of Cephalotaxus fortunei Hook.f is inducting juvenile stem segments in winter and culturing in medium MS,after the callus having been subcultured on the same medium for 6-8 generations,the callus with more alkaloids are obtained.
