Title: In Vitro Culture and Establishment of Regeneration System of Callistephus chinensis
Abstract: [Objective] The study aimed to establish the rapid propagation in vitro and regeneration system of Callistephus chinensis.[Method] With test-tube plantlets of C.chinensis through aseptic seeding as explants,MS basic medium was used as the media by adding different concn.of 6-BA,KT,NAA,the expanding propagation and rooting culture test were carried out on C.chinensis test-tube plantlets so as to research the effect of different growth regulator on subculture and rooting culture of C.chinensis test-tube plantlets and screen the optimal medium for rapid propagation and regeneration system of C.chinensis.[Result] 6-BA concn.had a significant influence on callus induction,and the induction rate was increased significantly with the increment of BA concn..KT concentration had the lowest influence on callus induction.The optimum medium for rapid propagation of C.chinensis was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L,with the meristematic number of 5.67±0.82 and the callus induction rate of(53.33±0.09)%;The best medium for regeneration system was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L,with the meristematic number of 6.33±1.03 and the callus induction rate of(83.70±0.04)%.When test-tube plantlets were cultured on the medium of MS+NAA 0.2 mg/L they got rooting rate of over 98.0% and the survival rate of over 90.0% after hardening and transplanting.[Conclusion] In the experiment,the rapid propagation in vitro and regeneration system for C.chinensis were established.
