Title: The effect of musclin for PI3K/AKT(PKB) pathway in the way of insulin-induced differented rat myocytes glucose uptakeing
Abstract: Objective: To study the signal pathway in which musclin inhibited insulin-stimulated glucose uptake.Methods:P3 differented rat myocytes were divided into four groups: control group(A,25 mmol·L-1 D-glucose) and musclin group(B,25 mmol·L-1 D-glucose+ 0.5 μg·mL-1 musclin)without insulin,control group(C,25 mmol·L-1 D-glucose+10 mU·mL-1 insulin) and musclin group(D,25 mmol·L-1 D-glucose+ 10 mU·mL-1 insulin +0.5 μg·mL-1 musclin)with insulin.B and D groups were incubated for 5h after adding musclin,and then C and D groups were incubated for another 30 min after adding insulin.Glucose uptake was assayed using 3H-G.The expression levels of GLUT4,PI3K and AKT mRNAs were measured by semiquantitative RT-PCR.Results: Compared with A group,B group was decreased by 50%(P0.01)in the ability of insulin-induce 3H-glucose(3H-G) uptake,and the expression levels of PI3K and AKT(PKB) mRNA.Compared with C group,D group was decreased by 50%(P0.01);Compared with B group,D group was increased about 30%(P0.05),while the expression level of GLUT4 had no significant difference among four groups.Conclusions: Musclin inhibited insulin-stimulated glucose uptake,and its mechanism maybe related to PI3K/AKT pathway.
