MethodsDiabetes ...ObjectiveTo investigate the effects of puerarin on renal structure, function and the expression of nitrotyrosine(NT) as the specific symbol of peroxynitrite (ONOO -) in diabetic rats. MethodsDiabetes mellitus models were established in rats by injecting streptozotoein (65 mg/kg) once intraperitoneally. The drug administration groups had daily injections of puerarin intraperitoneally for 16 weeks after induction of diabetes mellitus. The urine was collected for 24 hours as sample of urine protein and urine creatinine determination. Blood was sampled from the abdominal aorta, and serum was isolated for determination of blood glucose, blood creatinine and blood urea nitrogen. Pathological changes of the renal tissues were observed under light microscope. NT expression in renal tissues was detected by immunohistochemical technique. ResultsCompared with control group, abnormalities concerning kindey function as well as morphological changes such as hypertrophy of glomerulus and renal tubule were observed in diabetic rats, and NT expression was increased in diabetic kidney. Puerarin administration improved the kidney function and its pathological changes, and resulted in lower expression on NT in the kidney compared with those in diabetes mellitus group. ConclusionPuerarin brings down NT expression in the renal tissues and alleviates the oxidative injury to the kidney, which may act as a mechanism in alleviation of renal function and structure abnormities in rats by diabetes mellitus.Read More
