Abstract:Objective Observe the changes of renal function in the type 2 diabetic rats model induced by streptozotocin,and explore whether this model is suitable for the study of diabetes renal damage.Methods Di...Objective Observe the changes of renal function in the type 2 diabetic rats model induced by streptozotocin,and explore whether this model is suitable for the study of diabetes renal damage.Methods Diabetes mellitus animal models were induced by intraperitoneal injection of streptozotocin(STZ) and a high-fat diet.The diabetic rats are categorized as the diabetes group(DG,n = 10),the normal SD rats are categorized as the normal control group(NCG,n =8).The blood glucose,blood urea nitrogen,serum creatinine,serum AGE,urine microalbumin and kidney weight index were measured 8weeks after STZ injection in two groups.The expressions of RAGE,PKC and PKA in renal tissue were measured by Real Time PCR(RT— PCR) and Western blot.Results The blood glucose,blood urea nitrogen,serum creatinine,serum AGE,urine microalbumin and Kidney weight index level of DG group were significantly higher than NCG group(P 0.05).The mRNA and protein expression of RAGE and PKC of DG group were higher than NCG group(P 0.05).The mRNA and protein expression of PKA of DG group were significantly lower than NCG group.(P 0.05).Conclusion The type 2 diabetic rat model induced by STZ has significant damage of renal function in 8 weeks after STZ injection;The important upstream factors of kidney damage—RAGE,PKC and PKA were significantly changed,Therefore this animal model could be used for the study of diabetic renal damage and it's suitable for observation of early diabetic nephropathy.Read More
